  • Success of in vitro culture depends on the right composition of the culture medium.
  • Culture media contain inorganic components, organic supplements, carbon source, plant growth hormones, gelling agents, and antibiotics.
  • Macronutrients are required in milli molar concentrations and include carbon, calcium, hydrogen, potassium, magnesium, nitrogen, oxygen, phosphorus, and sulphur.

  • Plant tissue culture can be categorised as organ culture, callus culture, cell suspension culture, and protoplast culture.
  • Organ culture involves organs such as root, anther, ovary, embryo, endosperm, or seed under laboratory conditions.
  • Callus culture produces an unorganised mass of cells called callus.
