1. Basic idea and historical development
- Plant tissue culture is the cultivation of undifferentiated plant cells, tissues, or organs on artificial media under aseptic and controlled conditions.
- Totipotency is the basis of plant tissue culture.
- Schwann and Schleiden established that the cell is the basic unit of life capable of dividing and growing.
2. Explants, sterilisation, and basic steps of culture
- Virtually any plant part such as leaf, apical meristem, embryo, cotyledon, or hypocotyl can be used as an explant.
- Immature embryos are generally more responsive than apical meristems, and apical meristems are generally more responsive than leaf explants under suitable conditions.
- Explants from seedlings grown in sterile conditions do not need re-sterilisation.
3. Nutrient media and its components
- Success of in vitro culture depends on the right composition of the culture medium.
- Culture media contain inorganic components, organic supplements, carbon source, plant growth hormones, gelling agents, and antibiotics.
- Macronutrients are required in milli molar concentrations and include carbon, calcium, hydrogen, potassium, magnesium, nitrogen, oxygen, phosphorus, and sulphur.
4. Culture types and morphogenesis
- Plant tissue culture can be categorised as organ culture, callus culture, cell suspension culture, and protoplast culture.
- Organ culture involves organs such as root, anther, ovary, embryo, endosperm, or seed under laboratory conditions.
- Callus culture produces an unorganised mass of cells called callus.
