Topics in Host–Vector System

1

1. Core idea of host–vector system

  • Recombinant DNA technology works through a compatible host and a vector.
  • The desired DNA molecule is called the insert or target gene.
  • The carrier DNA molecule is called the vector.
2

2. Features of a good vector

  • A vector should be small in size.
  • A vector should have an origin of replication, or ori.
  • A vector should have unique restriction sites.
3

3. Plasmids as vectors

  • Plasmids are circular, double-stranded, extra chromosomal DNA capable of autonomous replication.
  • Plasmids occur naturally in bacteria, archaea, and yeast.
  • Plasmids can integrate into chromosomes; such plasmids are called episomes.
4

4. Development of plasmid vectors

  • pBR313 was an early plasmid-based vector and was 9 kb in size.
  • pBR313 carried tetR and ampR selectable markers.
  • pBR313 replicated in a relaxed fashion.

Key Concepts

Central concept: Recombinant DNA technology depends on a compatible host and a vector working together for cloning, propagation, and expression of foreign DNA.
