What to Focus On

  • A candidate gene is selected based on its biomedical, agricultural, or economic significance
  • A haploid human genome contains approximately 3.2 billion base pairs
  • A typical gene is only 3,000–3,500 bp — about one-millionth of the entire genome

  • Two major challenges in nucleic acid isolation: small quantities relative to other biomolecules, and susceptibility to physical/chemical damage
  • Four steps in nucleic acid isolation: cell disruption, protection from degrading enzymes, separation from other molecules, precipitation
  • Lysozyme digests bacterial cell wall; SDS disrupts cell membranes

  • Exonucleases remove nucleotides from the ends; endonucleases cleave internal bonds
  • Restriction enzymes recognise specific palindromic sequences of 4–8 bp
  • Type II restriction enzymes are used in rDNA technology as they cleave within the recognition sequence and need only Mg²⁺

Common Mistakes to Avoid

In blue-white selection, blue colonies contain recombinant DNA (with insert) and white colonies are non-recombinant.

Restriction enzymes cut DNA at random positions to generate fragments of different sizes.

All three types of restriction enzymes (Type I, II, III) are used in recombinant DNA technology.
